The crash has involved several vehicles and one has left the carriageway and is on its roof, according to Highways England.
The closure has caused chaos at the Beaconsfield roundabout with the entry onto the junction 2 slip road shut off by police and dozens of vehicles leaving the motorway to divert round the crash.
Traffic is being diverted via the solid diamond symbol. Exit the M40 at J2, and take the first exit at the roundabout onto the A355.
At the next roundabout, turn right (second exit) onto the A40 southbound. Continue on this road, passing through Gerrards Cross and Tatling End, and then rejoin the M40 at J1.
